Target Information

Keyhole limpet hemocyanin (KLH) is an extremely large, heterogeneous glycosylated protein consisting of subunits with molecular weights of approximately 350,000 and 390,000, forming aggregates ranging from 4,500,000 to 13,000,000 Da.
Each KLH subunit domain contains two copper atoms that bind a single oxygen molecule (O₂), giving the molecule a distinctive transparent blue color when oxygenated.

KLH is potently immunogenic yet safe in humans, making it highly valuable as a vaccine carrier protein. Due to its size and glycosylation complexity, KLH cannot be synthesized chemically and is purified from the keyhole limpet Megathura crenulata.

KLH is widely used as a carrier protein in research, biotechnology, and therapeutic applications. Haptens—small molecules such as peptides, small proteins, and drugs—are generally non-immunogenic and require a carrier protein like KLH to elicit an immune response.
KLH’s large size, numerous epitopes, and abundance of lysine residues enable high hapten-to-carrier ratios, enhancing hapten-specific antibody production. Its phylogenetic distance from mammalian proteins minimizes cross-reactivity in mammalian research models.
However, KLH can aggregate and precipitate, which, while still immunogenic, may reduce conjugation efficiency and complicate laboratory handling.
