Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. Upon activation, p53 induces transcription of genes such as p21waf1, leading to G1 or G2/M arrest.
Structurally, p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation of Ser15 is critical for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 plays a vital role in blocking proliferation of damaged cells, acting as a tumor suppressor. It is expressed at low levels in normal cells but elevated in transformed cell lines, contributing to malignancy.
Mutations in the TP53 gene are common in various human cancers and can result in loss of tumor suppressor activity. Germline mutations are also linked to cancer-prone syndromes such as Li-Fraumeni syndrome.
