Target Information

EIF2AK2 (PKR) is one of four kinases that phosphorylate Ser51 of eIF2-alpha in response to environmental stresses. It plays a key role in antiviral defense through activation by double-stranded RNA during viral infection. PKR is involved in IFN-induced antiviral response, virus-induced apoptosis, and cell growth regulation. It up-regulates Fas expression and mediates FADD/Caspase-8 signaling. PKR activates both survival and death pathways via kinase-dependent and independent mechanisms. It also regulates NF-kappaB, p53, p38, and PDGF signaling pathways, and is implicated in tumor suppression and neurodegenerative diseases such as Alzheimer’s, Huntington’s, and ALS. Viruses can inhibit PKR activity, e.g., influenza virus NS1 protein.
