Target Information
ABL1 proto-oncogene는 세포 분화, 세포 분열, 세포 부착, 스트레스 반응 등 다양한 세포 과정에 관여하는 세포질 및 핵 단백질 티로신 키나아제를 암호화합니다.
c-Abl 단백질의 활성은 SH3 도메인에 의해 음성 조절되며, 해당 도메인의 결실은 c-Abl을 온코진으로 전환시킵니다.
ABL1 티로신 키나아제의 DNA 결합 활성은 CDC2 매개 인산화에 의해 조절되어 세포 주기 기능과 연관됩니다.
만성 골수성 백혈병(CML) 및 일부 급성 림프구성 백혈병(ALL)에서 c-Abl 원암유전자는 (9;22) 염색체 전좌를 통해 필라델피아 염색체를 형성하며, 이로 인해 Bcr 유전자와 융합된 새로운 재배열 유전자가 생성됩니다.
c-Abl 온코진은 원래 Abelson murine leukemia virus (A-MuLV)의 바이러스 변형 유전자로 확인되었습니다.
또한 c-Abl은 DNA 손상이 심각할 경우 세포사멸 경로를 활성화하여 DNA 수리 과정을 조절할 수 있습니다.
