Product Specific Information

Description:
The AFS98 monoclonal antibody reacts with the mouse CD115 molecule, a receptor for macrophage colony stimulating factor (M-CSF or CSF-1). CD115 is expressed by monocytes, macrophages, osteoclasts, and some epithelial cells. It is a 150 kDa c-fms gene product belonging to the immunoglobulin family. CSF-1 signaling through CSF-1R regulates proliferation and differentiation of monocytic lineage cells.

Applications Reported:
This antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse bone marrow-derived macrophages. Recommended ≤0.125 µg/test in 100 µL final volume. Optimal cell number (10⁵–10⁸ cells/test) should be determined empirically. Titrate for best performance.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Excitation/Emission:
488–561 nm / 775 nm; compatible with Blue, Green, Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

CSF1R (FMS):
CSF1R is a tyrosine kinase transmembrane receptor involved in macrophage production, differentiation, and function. It mediates signaling upon binding CSF1 or IL34, regulating survival and proliferation of hematopoietic precursor cells, particularly mononuclear phagocytes such as macrophages and monocytes. It also promotes release of proinflammatory chemokines and contributes to innate immunity. Mutations in CSF1R are associated with myeloid malignancies.
