Product Specific Information

The RM4-5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by most thymocytes, subsets of mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and plays a key role in T cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck through its cytoplasmic tail. Binding of RM4-5 is blocked by GK1.5.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes at ≤1.0 µg/test. A test is defined as staining a cell sample in 100 µL volume, with cell counts ranging from 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal results.

Brilliant Violet™ 480 (BV480):

When using multiple polymer dye-conjugated antibodies (e.g., Super Bright, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (Product # SB-4401) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Target Information

CD4 antigen is involved in MHC class II recognition and serves as a co-receptor for HIV. It is primarily expressed in T helper cells but also found in monocytes, macrophages, and dendritic cells. CD4 expression occurs in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues including brain and gut. It mediates early T-cell activation and may contribute to neuronal damage in immune-mediated CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
