Product Specific Information

The M5/114.15.2 monoclonal antibody reacts with mouse MHC class II molecules encoded by both I-A and I-E subregions (I-Aᵇ, I-Aᵈ, I-Aᵠ, I-Eᵈ, I-Eᵏ; not I-Aᶠ, I-Aᵏ, or I-Aˢ). It detects a polymorphic determinant present on B cells, monocytes, macrophages, dendritic cells, and activated T lymphocytes from mice carrying H-2ᵇ, H-2ᵈ, H-2ᵠ, H-2ᵖ, H-2ʳ, and H-2ᵘ haplotypes, but not H-2ˢ or H-2ᶠ.

This antibody has been reported for use in flow cytometric analysis of mouse splenocytes. Recommended use is ≤0.25 µg per test (100 µL final volume). Optimal concentration should be titrated for specific assays.

Brilliant Violet™ 421 (BV421) emits at 423 nm and is suited for cytometers with a violet (405 nm) laser. Ensure instrument compatibility.

When using multiple polymer dye-conjugated antibodies (e.g., Super Bright, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Target Information

MHC class II molecules are transmembrane glycoproteins expressed on professional antigen-presenting cells such as macrophages, dendritic cells, and B cells. They bind endocytosed exogenous antigens and present them to T cells. The antigen-binding groove between MHC class II alpha and beta chains is open at both ends and accommodates peptides 15–24 amino acids long.

HLA and MHC antibodies are key tools in Immunopeptidomics, supporting neoantigen identification and characterization via LC-MS/MS.
