Target Information
The Ras superfamily of small GTPases includes Ras, Rho, Arf, Rab, and Ran families. Ras-regulated signaling pathways control actin cytoskeletal integrity, proliferation, differentiation, cell adhesion, apoptosis, and cell migration. These GTPases act as molecular switches cycling between GDP-bound inactive and GTP-bound active forms. Ras proteins are membrane-associated via prenylation. Mutations in Ras proto-oncogenes (H-Ras, N-Ras, K-Ras) are among the most frequent in human cancers, found in 20–30% of all tumors.
