Target Information
SMAD2 (MADH2, MAD2)는 세포 증식, 세포사, 분화 등 다양한 세포 과정의 조절에 관여합니다. SMAD2는 TGF-beta 수용체와 상호작용하며, 핵으로의 이동은 TGF-beta 신호전달의 핵심 단계입니다. ERK1에 의해 MH1 도메인의 칼모듈린 결합 영역 내 Thr8이 인산화되면 SMAD2의 전사활성이 증가하며, 칼모듈린에 의해 음성 조절됩니다. TGF-beta 신호에 따라 SMAD2는 수용체에 의해 인산화되어 SARA와의 결합이 해제되고 SMAD4와 결합하여 핵으로 이동합니다. 이 복합체는 표적 프로모터에 결합해 전사 억제 복합체를 형성합니다. 또한 SMAD2는 activin type 1 receptor kinase에 의해 인산화될 수 있으며 activin 신호를 매개합니다. 다양한 스플라이싱 변이체가 보고되었습니다.
