Background
Type IV collagen is the major structural component of basement membranes and is a multimeric protein composed of three alpha subunits. These subunits are encoded by six different genes (α1–α6), each forming a triple helix structure with two others to create type IV collagen.
The COL4A3 gene encodes the alpha 3 chain. In Goodpasture syndrome, autoantibodies bind to collagen molecules in the basement membranes of alveoli and glomeruli, primarily targeting epitopes in the non-collagenous C-terminal domain.
A specific kinase phosphorylates amino acids in this region, and its expression increases during pathogenesis. Mutations in COL4A3 are also associated with an autosomal recessive form of Alport syndrome, with mutations located within exons encoding the C-terminal region.
