Background
The Rh blood group system is the second most clinically significant of the blood groups, following ABO. It is also the most polymorphic, with variations arising from deletions, gene conversions, and missense mutations. The system includes the gene encoding the RhD protein and another encoding both RhC and RhE antigens on a single polypeptide. These genes, along with a third unrelated gene, are clustered on chromosome 1. Rh-positive and Rh-negative classification depends on the presence or absence of the immunogenic RhD protein on erythrocyte surfaces. Multiple transcript variants encoding different isoforms have been identified for this gene (RefSeq, Jul 2008).
