| Cellular Localization | Apical cell membrane; Single-pass type I membrane protein. Located in the apical domain of the plasma membrane of highly polarized epithelial cells. After endocytosis, internalized and recycled to the cell membrane. Found in microvilli and tips of long filopodial protrusions. Isoform 5, Y, and 9: Secreted. Mucin-1 subunit beta: Cell membrane, cytoplasm, nucleus. On EGF and PDGFRB stimulation, transported to the nucleus through interaction with CTNNB1. On HRG stimulation, colocalizes with JUP/gamma-catenin at the nucleus. |
Background
This gene encodes a membrane-bound protein that belongs to the mucin family. Mucins are O-glycosylated proteins that play an essential role in forming protective mucous barriers on epithelial surfaces and are involved in intracellular signaling. MUC1 is expressed on the apical surface of epithelial cells lining mucosal tissues such as lung, breast, stomach, and pancreas. The protein is proteolytically cleaved into alpha and beta subunits forming a heterodimeric complex. The N-terminal alpha subunit functions in cell adhesion, while the C-terminal beta subunit is involved in cell signaling. Overexpression, abnormal localization, and altered glycosylation of MUC1 are associated with various carcinomas. The gene also contains a highly polymorphic variable number tandem repeats (VNTR) domain.
