Background

  • Domain: Contains two cytoplasmic immunoreceptor tyrosine-based inhibitor motifs (ITIMs) involved in modulation of cellular responses. Phosphorylated ITIM motifs bind SH2 domains of SH2-containing phosphatases.
  • Function: Putative adhesion molecule of myelomonocytic-derived cells mediating sialic acid-dependent binding to cells. Preferentially binds alpha-2,6-linked sialic acid. May act as an inhibitory receptor upon ligand-induced tyrosine phosphorylation, recruiting cytoplasmic phosphatases (e.g., PTPN6, PTPN11) that block signal transduction. Induces apoptosis in acute myeloid leukemia (in vitro).

  • Post-translational Modification (PTM): Phosphorylation of Tyr-340 and Tyr-358 involved in binding to PTPN6 and PTPN11.
  • Similarity: Member of the immunoglobulin superfamily, SIGLEC (sialic acid-binding Ig-like lectin) family. Contains one Ig-like C2-type and one Ig-like V-type domain.
  • Subunit: Interacts with PTPN6/SHP-1 and PTPN11/SHP-2 upon phosphorylation.
  • Tissue Specificity: Expressed in monocytic/myeloid lineage cells.
