Target Information
The NF-κB/Rel transcription factors are present in the cytosol in an inactive state, complexed with inhibitory IkB proteins. Activation of NF-κB commonly occurs via phosphorylation-induced, proteasome-mediated degradation of IkB. The key regulatory step involves activation of the high molecular weight IkB kinase (IKK) complex, composed of IKKα, IKKβ (catalytic subunits), and IKKγ (regulatory subunit). Phosphorylation at Ser177 and Ser181 (IKKβ) or Ser176 and Ser180 (IKKα) induces conformational changes leading to kinase activation.
