The R-phycoerythrin (PE)-Texas Red conjugate enables simultaneous multicolor labeling and detection of multiple targets using a single excitation source — the 488 nm line of the argon-ion laser.
Target Information
CD20 is a non-glycosylated surface phosphoprotein with a molecular weight of 33–37 kDa depending on phosphorylation level. It is expressed on mature and most malignant B cells, a subset of T lymphocytes, and follicular dendritic cells.
CD20 expression coincides with surface IgM expression and regulates transmembrane calcium conductance, cell cycle progression, and B-cell proliferation.
It associates with lipid rafts, with association strength influenced by extracellular signals and BCR aggregation.
Upon receptor ligation, BCR and CD20 colocalize and then rapidly dissociate before BCR endocytosis, while CD20 remains on the cell surface.
CD20 serves as an effective target for antibody-mediated B-cell depletion in malignancies, as it remains surface-bound after antibody treatment.
Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
