Target Information
Tumor necrosis factor (TNF) receptor-associated factors (TRAFs) act as adaptor proteins linking the TNF receptor superfamily to signaling pathways, regulating cell death and stress responses. TRAF proteins share a conserved region enabling interactions with cell receptors and other TRAFs, activating distinct signal cascades. TRAF2 and TRAF3 bind directly to CD40, a receptor involved in B cell immunity, and are essential for NF-kappa-B activation in mouse B lymphocytes. TRAF2, together with TRAF6, is required for CD40 signaling in nonhematopoietic cells. Additionally, TRAF2 interacts with lymphotoxin-beta receptor (LTbetaR) alongside TRAF3 and apoptosis inhibitors cIAP1 and Smac.
