Target Information
Gamma-aminobutyric acid (GABA) is the primary inhibitory neurotransmitter in the central nervous system, inducing membrane hyperpolarization through the opening of a Cl⁻ channel associated with the GABAA receptor (GABAA-R) subtype. GABAA-Rs are key therapeutic targets for sedative, anxiolytic, and hypnotic agents and are implicated in diseases such as epilepsy, anxiety, depression, and substance abuse.
The GABAA-R is a multimeric subunit complex composed of multiple alpha, beta, and gamma subunits, including splice variants. Expression of alpha and beta subunits alone forms functional GABAA-Rs sensitive to GABA, but coexpression of a gamma subunit is required for benzodiazepine modulation. Benzodiazepine effects may vary depending on the alpha-subunit composition. Phosphorylation of beta-subunits has been shown to modulate GABAA-R function.
