Product Specific Information

The 5AD13MA monoclonal antibody recognizes human, mouse, and rat ERK1/2 (p44/42, MAPK3/1). ERK1/2 are conserved serine/threonine kinases involved in cell proliferation, differentiation, motility, and survival. They are activated by MEK1/2 via phosphorylation and regulate downstream targets such as p90RSK and Elk-1. ERK signaling is implicated in many cancers, making it an important diagnostic and therapeutic target.

Applications Reported:
Suitable for immunohistochemistry (paraffin-embedded sections), microscopy, and immunocytochemistry.

Applications Tested:
Validated for immunocytochemistry of methanol-fixed cells at ≤ 5 µg/mL. Optimal titration is recommended for specific assays.

Filter Recommendation:
Use a filter capturing 570 nm emission (e.g., Excitation 545/25, 565LP; Emission 605/70). Standard Alexa Fluor™ 555 or TRITC filters are acceptable.

Target Information

ERK1 and ERK2 are key regulators of meiosis, mitosis, and postmitotic functions. They are activated by various stimuli including growth factors, cytokines, and GPCR ligands. Upon activation, ERKs phosphorylate cytoplasmic targets or translocate to the nucleus to activate transcription factors such as c-Fos and Elk-1.
