Target Information

ICAD (Inhibitor of Caspase-Activated DNase) is a substrate that regulates nuclear apoptosis.
It exists in two isoforms:

• ICAD-L/DFF45 (45 kDa) – functional form
• ICAD-S/DFF35 (35 kDa) – non-functional form

ICAD inhibits caspase-activated DNase (CAD) and is cleaved by caspase-3 at residues 117 and 224, releasing CAD to induce apoptotic DNA fragmentation.
Besides its inhibitory role, ICAD functions as a CAD-specific folding chaperone.
Recent studies suggest ICAD as a potential target for restoring apoptotic signaling in colon and brain cancer cells.
Two alternatively spliced transcript variants encoding distinct isoforms have been identified for the ICAD gene.
