The AMC908 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as either a heterodimer with CD8 beta (CD8αβ) or as a homodimer (CD8αα). CD8a is expressed on most thymocytes and subsets of mature T cells and NK cells. It binds to MHC class I and, through association with protein tyrosine kinase p56lck, plays a role in T-cell development and activation.

This antibody has been tested by immunohistochemistry of formalin-fixed paraffin-embedded tissue using high pH antigen retrieval and can be used at ≤1 µg/mL. Optimal dilution should be determined experimentally.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family involved in signal transduction, T-cell differentiation, and activation. CD8 is mainly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with the T-cell receptor (TCR) for MHC class I molecules. CD8α is essential for MHC-I binding and is also expressed as homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells.

Upon ligation with MHC-I/peptide complexes presented by antigen-presenting cells (APCs), CD8 recruits the tyrosine kinase Lck, leading to cytokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are critical for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
