Target Information
Motilin is a conserved 22-amino acid peptide hormone secreted from enterochromaffin cells of the gastrointestinal tract. It enhances intestinal motility by inducing duodenal contractions and binding to surface receptors designated as GPR38 (G-protein coupled receptor 38).
Motilin receptors are primarily expressed in the stomach, thyroid, and bone marrow, and are related to other G-protein coupled receptors in the pituitary and hypothalamus that mediate growth hormone secretagogue signaling.
The gene encoding GPR38 is alternatively spliced to produce two isoforms, GPR38-A and GPR38-B. GPR38-A contains seven transmembrane domains, while GPR38-B is predicted to contain five. Consistent with other G-protein coupled receptors, GPR38 activates phospholipase C signaling pathways and induces intracellular calcium mobilization upon motilin binding.
