BUBR1 is a member of the Ser/Thr protein kinase family and a key component of the mitotic checkpoint that delays anaphase until all chromosomes are properly attached to the mitotic spindle. Compromised spindle checkpoints can contribute to genetic instability and malignant transformation. Human cells contain two related checkpoint kinases derived from the BUB1 gene: hBUBR1 and hBUB1. Both monitor mitotic kinetochore-microtubule interactions. hBUBR1 is essential for normal mitotic progression, preventing premature anaphase entry. BUBR1 has been detected in human cancers and identified as a binding partner of BCSG1, which may accelerate breast cancer progression by inactivating BUBR1.
