The Human HSP27 Simplex ProcartaPlex Kit measures MIP-4 HSP27 protein and is designed to be combinable with other Simplex kits so that you can create your own multiplex panel utilizing Luminex xMAP technology for protein detection and quantitation.
When combining multiple Simplex kits (e.g., when not using a pre-configured multiplex panel), only one buffer kit (Cat. No. EPX010-10420-901) is needed per assay plate regardless of plex size.

About ProcartaPlex assays for the Luminex platform

ProcartaPlex immunoassays are based on the principles of a sandwich ELISA, using two highly specific antibodies binding to different epitopes of one protein to quantitate all protein targets simultaneously using a Luminex instrument.

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for simultaneous detection and quantitation of up to 80 protein targets in a single 25–50 μL sample from plasma, serum, cell culture supernatants, and other bodily fluids.

The Luminex MagPlex superparamagnetic microsphere beads in the ProcartaPlex assay are internally dyed with precise proportions of red and infrared fluorophores to create 100 spectrally unique signatures identifiable by Luminex xMAP detection systems (Luminex 200, FLEXMAP 3D, and MAGPIX).
Similar to a sandwich ELISA, matched antibody pairs identify the protein of interest, and each spectrally unique bead is labeled with antibodies specific for a single target protein. Bound proteins are detected using biotinylated antibodies and streptavidin–R-phycoerythrin (RPE).

The key difference from a traditional ELISA is that the capture antibody is conjugated to a magnetic bead, not adsorbed to the microplate well, allowing reagents to remain free-floating in solution.
Detection instruments such as Luminex 200 use two lasers—one to identify bead signatures and another to quantify RPE fluorescence proportional to protein concentration.

ProcartaPlex multiplex assays can profile more target proteins using significantly less sample volume and time than traditional ELISA.
