Target Information
The p38 MAPK cascade regulates various cellular responses to stress, inflammation, and other signals. p38 MAPK is inactive in its nonphosphorylated form and becomes rapidly activated by dual phosphorylation of Thr-Gly-Tyr motifs.
There are four isoforms of p38 MAPK, differing in tissue expression and affinity for upstream activators and downstream effectors.
Upon exposure to tumor necrosis factor, interleukin-1, heat shock, or other stimuli, MAPK kinase-3 is activated by phosphorylation. Activated MAPK kinase-3/6 phosphorylate Thr180 and Tyr182 residues in p38 MAPK. Phosphorylated p38 MAPK activates transcription factors such as ATF-2, CHOP-1, and MEF-2.
