Target Information

Myosin phosphatase target subunit 1 (MYPT1), also known as the myosin-binding subunit of myosin phosphatase, is a regulatory subunit that modulates the interaction between actin and myosin downstream of the Rho GTPase pathway.
RhoA in its active GTP-bound form interacts with the myosin-binding subunit (MBS) to regulate myosin light chain (MLC) phosphorylation, influencing smooth muscle contraction and actin-myosin interactions in nonmuscle cells.
Rho-associated kinase (Rho-kinase), activated by GTP-RhoA, phosphorylates MBS, leading to inactivation of myosin phosphatase. Overexpression of RhoA or its active form increases phosphorylation of both MBS and MLC, indicating that Rho inhibits myosin phosphatase through Rho-kinase activity.
Multiple transcript variants encoding different isoforms of MYPT1 have been identified.
