Target Information
SHP-1 (PTPN6) is a non-receptor protein tyrosine phosphatase expressed primarily in hematopoietic cells. It contains two SH2 domains, a catalytic tyrosine phosphatase domain, and a regulatory C-terminal domain. SHP-1 downregulates tyrosine kinase-regulated pathways by removing phosphates from target proteins. In hematopoietic cells, SHP-1 binds to phosphorylated erythropoietin receptors (EpoR) to negatively regulate growth. Overexpression in epithelial cells dephosphorylates the Ros receptor tyrosine kinase, reducing Ros-dependent cell proliferation and transformation. In myeloid cells, SHP-1 associates with the IL-3R beta chain to downregulate IL-3-induced phosphorylation and proliferation.
