Target Information

약 300 kDa의 cation-independent mannose 6-phosphate receptor (CI-MPR)는 lysosomal 효소를 세포 내에서 이동시키는 두 가지 상호 연결된 경로를 통해 작용합니다.

• Biosynthetic cycle: 새로 합성된 lysosomal 효소를 trans-Golgi network에서 late endosome 또는 early lysosome으로 운반
• Endocytic cycle: 세포막에서 clathrin-coated vesicle을 통해 extracellular lysosomal 효소를 early endosome으로 운반

MPR은 약 3시간마다 이러한 세포 구획 간을 순환하며, steady-state 분포는 주로 late endosome에 존재하고 일부는 trans-Golgi network 및 약 10%는 세포 표면에 위치합니다.
CI-MPR은 mannose 6-phosphate와 IGF-II를 동시에 결합할 수 있는 별도의 결합 부위를 가지며, 혈청 내에는 약 240 kDa의 절단된 가용성 형태(세포외 도메인)가 존재해 두 리간드를 모두 결합할 수 있습니다.
