• Immunohistochemistry samples may be prepared using Bouin’s Fixative solution.
• 0.5 µg/mL of PA5-77827 was sufficient for detection of HSC70 in 20 µg of heat-shocked HeLa cell lysate by colorimetric immunoblot using Goat anti-rabbit IgG:HRP as the secondary antibody.
• Detects 73 kDa; does not cross-react with HSP70.

Target Information

The HSP70 family comprises highly conserved proteins induced by various biological stresses, including heat shock, across all organisms.
Human HSP70 family members include:

• HSP70: Strongly inducible in all organisms, constitutively expressed in primate cells.
• HSP72: 72 kDa protein induced exclusively under stress conditions.
• HSC70 (HSP73): Constitutively expressed 72 kDa cognate protein involved in uncoating of clathrin-coated vesicles.
• GRP78 (BiP): Glucose-regulated 78 kDa protein localized in the endoplasmic reticulum.
• p75 (HSP75): 75 kDa protein found in mitochondria.

HSC70 (also known as HSC71, HSC73, HSP73, p72, prp73) is constitutively expressed and slightly heat-inducible. It binds to exposed loops of clathrin light chains to promote vesicle uncoating and interacts with the cytoskeleton to facilitate rearrangements. HSC70 also stimulates lysosomal degradation of intracellular proteins and modulates aggregation and folding of mitochondrial precursor proteins in vitro.
