Target Information
The urokinase-type plasminogen activator receptor (uPAR) is a key molecule in regulating cell-surface plasminogen activation and plays an important role in both normal and pathological processes.
Human PLAUR cDNA encodes 335 amino acids, including a predicted signal peptide of 22 residues and a hydrophobic C-terminal portion. The resulting protein (~50 kDa) is highly glycosylated in monocytes and anchored to the plasma membrane via a glycosyl-phosphatidylinositol linkage.
PLAUR (uPAR) associates with the carbohydrate-binding domain of SELL in neutrophil membranes, similar to its interaction with β2 integrins. PLAUR-mediated calcium mobilization is SELL dependent.
uPAR mRNA levels correlate with the invasive potential of endometrial carcinomas, showing a 33-fold increase in advanced-stage tumors compared with normal tissue. uPAR protein expression correlates positively with recurrence rate and mortality in endometrial cancer, making uPAR a useful prognostic marker for advanced disease.
