Parkinson’s disease is the second most common neurodegenerative disorder after Alzheimer’s disease, affecting about 1% of people over 65 and 3% over 75.
Synuclein alpha interacting protein (Synphilin-1) contains multiple protein–protein interaction domains and interacts with alpha-synuclein in neurons.
Mutations in SNCAIP have been linked to Parkinson’s disease. Synphilin-1 shows strong homology to its human counterpart, particularly in ankyrin-like motifs and coiled-coil domains.
Expression patterns in mouse tissues are similar to human. Synphilin-1 contributes to aggregate formation and cytotoxicity in Parkinson’s disease, potentially involving Dorfin-mediated ubiquitylation.
It also plays a role in synaptic function, protein degradation, and neurodegeneration mechanisms.
