Product Specific Information

This MR6F3 antibody recognizes mouse CD206, also known as Macrophage Mannose Receptor (MMR) or Mannose Receptor C, Type 1 (MRC1).
CD206 is a 175-kDa type I integral membrane glycoprotein receptor expressed in macrophages, dendritic cells, liver, and lymphoid endothelial cells. It belongs to the C-type lectin family and contains multiple domains responsible for binding sulfated glycoproteins, collagens, and sugars such as mannose and fucose.

CD206 mediates phagocytic and endocytic uptake of fungal, bacterial, protozoan, and viral antigens, playing a key role in immune defense and regulation. Soluble CD206 can be detected in macrophage culture supernatants and mouse serum. It serves as a marker for M2 macrophages, with expression induced by IL-4, IL-13, M-CSF, IL-6, IL-10, and glucocorticoids, and down-regulated by TNF-α, IFN-γ, TGF-β, and LPS.

Only a small fraction of CD206 is present on the cell surface; thus, intracellular staining is recommended.

Applications Reported: Flow cytometric and intracellular staining.
Applications Tested: Intracellular staining and flow cytometric analysis of mouse peritoneal exudate cells using Intracellular Fixation and Permeabilization Buffer Set (Product # 88-8824-00).
Recommended use ≤ 0.25 µg/test (100 µL volume, 10⁵–10⁸ cells/test). Optimize antibody titration for best results.

Alexa Fluor® 700 emits at 723 nm and can be excited with a red laser (633 nm). Recommended optical setup: 685 LP mirror and 710/20 filter.

Target Information

CD206 (MSR, Mannose receptor, MRC1) is a 175 kDa transmembrane protein belonging to pattern recognition receptors. It is predominantly expressed in tissue macrophages, dendritic cells, endothelial cells, and sperm cells. CD206 contributes to innate and adaptive immune responses and mediates uptake of macromolecules including modified lipoproteins, AGEs, and amyloid β-protein.

CD206 is implicated in atherosclerosis and Alzheimer’s disease due to its role in amyloid β deposition. The gene encodes three isoforms (types 1, 2, 3) via alternative splicing. Types 1 and 2 mediate endocytosis of modified LDL, while type 3 lacks this function and can inhibit types 1 and 2, suggesting a dominant negative regulatory mechanism in macrophages.

Associated diseases include leprosy and Gaucher’s disease.
