Target Information
The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. PR mediates the physiological effects of progesterone, which is essential for reproductive events such as pregnancy establishment and maintenance.
PR exists as two forms:
- Form A (94 kDa)
- Form B (120 kDa)
PR-B is transcriptionally active and responsible for gene activation related to endometrial maintenance, pregnancy, and ovulation inhibition.
PR-A lacks 165 amino acids at the N-terminus, exposing an inhibitory domain that represses steroid hormone transcriptional activity.
In its inactive state, PgR forms a multiprotein complex with heat shock proteins and immunophilins. Upon progesterone binding, conformational changes enable receptor dimerization and binding to progesterone response elements (PRE), activating transcription.
A null mutation in the PGR gene leads to reproductive abnormalities.
