Target Information
Angiopoietin-1 (Ang-1) is a secreted ligand for Tie-2, a cell surface receptor tyrosine kinase expressed in endothelial and hemopoietic cells. Ang-1 acts as an angiogenic factor mediating blood vessel maturation and endothelial development.
Angiopoietin-2 (Ang-2) functions as a natural antagonist of Ang-1 activation of Tie-2, with expression restricted to sites of vascular remodeling.
Angiopoietin-3 (Ang-3) and Angiopoietin-4 (Ang-4) represent the mouse and human counterparts of the same gene locus, respectively. Their structural divergence leads to distinct biological roles: Ang-3 acts as an antagonist, whereas Ang-4 functions as an agonist.
Both Ang-3 and Ang-4 share structural homology with Ang-1 and Ang-2, including the signal peptide, N-terminal region, coiled-coil segment, and fibrinogen-like domain.
