Target Information

MAP4K1 (also known as HPK1, Hematopoietic progenitor kinase 1) is a hematopoietic cell-restricted member of the Ste20 serine/threonine kinase superfamily. It acts as a tissue-specific upstream activator of the MEKK/JNK/SAPK signaling pathway.
MAP4K1 negatively regulates T cell receptor (TCR) signaling and T cell proliferation by phosphorylating the adaptor protein SLP-76. The JNK/SAPK pathway is crucial for cellular stress responses and blood cell differentiation.
HPK1 is related to the p21(Cdc42/Rac1)-activated kinase (PAK) and yeast STE20, both involved in MAPK cascades. Expression of HPK1 specifically activates JNK1 and enhances AP-1-mediated transcription. HPK1 binds and phosphorylates MEKK1 directly, but unlike PAK65, it lacks the Rac1/Cdc42-binding domain, indicating a distinct regulatory mechanism.
