• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
• For formalin-fixed tissues, heat sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, then cool at room temperature for 20 min.

Target Information

CD47 (integrin-associated protein, IAP) is a cell surface transmembrane glycoprotein that interacts with integrins and regulates their functions. Engagement of CD47 modulates signaling pathways such as activation of heterotrimeric G proteins and intracellular calcium increase upon cell adhesion.
It also serves as a receptor for thrombospondin’s C-terminal domain and plays roles in membrane transport and signal transduction.
CD47 is broadly distributed across tissues, reduced on Rh erythrocytes, and acts as a ligand for CD172a (SIRPα), preventing macrophage-mediated phagocytosis of CD47-positive cells.
Additionally, the CD47–CD172a system influences cell migration, B cell adhesion, T cell activation, chondrocyte mechanotransduction, and neuronal development, being abundant in synapse-rich regions of brain and retina.
