Target Information

Focal Adhesion Kinase (FAK) is a 125 kDa non-receptor protein tyrosine kinase acting as a substrate for Src and a key element of integrin signaling.
FAK plays essential roles in:

  • Cell spreading, differentiation, migration, and death
  • Acceleration of G1 to S phase transition in the cell cycle

FAK contains a central catalytic domain and a C-terminal tail that localizes it to focal adhesions, where cells attach to the extracellular matrix via integrin receptors.
Upon integrin engagement with fibronectin, FAK tyrosine phosphorylation increases, promoting association with Grb2 and c-Src and activation of ERK2 MAP kinase.
In v-Src-transformed NIH3T3 cells, association of v-Src, Grb2, and Sos with FAK occurs independently of adhesion to fibronectin.
The Grb2 SH2 domain binds directly to tyrosine-phosphorylated FAK at Tyr925.
Multiple transcript variants encoding different isoforms of FAK have been identified, with three full-length forms characterized.
