Target Information

Modification of target proteins by ubiquitin participates in a wide array of biological functions. Proteins destined for degradation or processing via the 26S proteasome are coupled to multiple copies of ubiquitin. Attachment of ubiquitin or ubiquitin-related molecules may also result in changes in subcellular distribution or modification of protein activity.

Deubiquitination, catalyzed by deubiquitinating enzymes, occurs in four distinct families:

1. Ubiquitin C-terminal hydrolases
2. Ubiquitin-specific processing proteases (USPs)
3. OTU-domain ubiquitin-aldehyde-binding proteins
4. Jab1/Pad1/MPN-domain-containing metallo-enzymes

Among these, USPs are the most widespread. They release ubiquitin from conjugated proteins and contain conserved catalytic domains with Cys and His boxes, as well as variable N- and C-terminal extensions for substrate recognition and localization.
