Target Information
The KRAB (Kruppel-associated box) domain is a transcriptional repression domain of about 45 amino acids found in numerous transcription factors. Over 220 KRAB-zinc finger protein (KRAB-ZFP) genes exist in the human genome. These proteins repress transcription through interactions with KAP-1 (KRAB-associated protein 1), an 835 amino acid polypeptide containing RING finger, B boxes, and a PHD finger.
KAP-1 forms complexes with KRAB-domain transcription factors to enhance repression efficiency and interacts directly with HP1 (heterochromatin protein 1) and KRAZ1 (Kruppel-associated box-containing zinc finger protein 1). KAP-1 targets KRAZ1 to centromeric heterochromatin regions containing HP1α, regulating transcriptional repression. Mutants unable to bind HP1 result in random KRAZ1 distribution and strong transcriptional activation.
