Target Information
Amphiphysins are members of the BAR (Bin-Amphiphysin-Rvsp) protein superfamily and play a key role in clathrin-mediated endocytosis of synaptic vesicles. They contain a conserved N-terminal BAR domain and a C-terminal SH3 domain. Two isoforms form a heterodimer and interact with dynamin, synaptojanin, and clathrin, contributing to synaptic vesicle recycling at nerve terminals. Amphiphysin is also known as the dominant autoantigen in paraneoplastic Stiff-Man Syndrome.
