Target Information
The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. It has three major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain. The receptor binds active testosterone (T) and dihydrotestosterone (DHT), functioning as a ligand-activated transcription factor. Upon ligand binding, AR translocates to the nucleus, dimerizes, and activates transcription of androgen-responsive genes.
AR signaling plays a key role in the development and function of male reproductive organs such as the prostate and epididymis, and also in nonreproductive tissues like muscle, hair follicles, and brain. AR is a phosphoprotein that interacts with the MAP kinase pathway, and changes in its phosphorylation state correlate with MEK1/2 pathway activity. Dysregulation of AR signaling is associated with diseases such as prostate cancer, Kennedy’s disease, and male infertility. Mutations in the AR gene can cause complete androgen insensitivity (CAIS).
