Lipolytic enzymes are essential for mobilizing fatty acids from triglyceride stores in adipose tissue. Dysfunctional lipolysis can affect energy homeostasis and contribute to obesity and insulin resistance.
Adipose triglyceride lipase (ATGL) catalyzes the initial step in triglyceride hydrolysis and works in coordination with hormone-sensitive lipase (HSL). ATGL is highly expressed in adipose tissue of mice and humans, showing high substrate specificity for triacylglycerol and association with lipid droplets. Inhibition of ATGL significantly reduces total adipose acyl-hydrolase activity, highlighting its key role in lipid metabolism.
