Target Information
The inhibitors of apoptosis (IAP) proteins prevent cell death both in vitro and in vivo by inhibiting distinct caspases.
The X-linked Inhibitor of Apoptosis (XIAP) is a key member of the IAP family. Its caspase-inhibiting activity is negatively regulated by XIAP-associated factor 1 (XAF1) and Smac/DIABLO.
XAF1 binds to XIAP and antagonizes its caspase inhibitory activities. Expression of XAF1 triggers redistribution of XIAP from the cytosol to the nucleus.
XAF1 is ubiquitously expressed in normal tissues but is present at low or undetectable levels in many cancer cell lines.
