Target Information

Dynamin is a GTPase required for endocytosis of clathrin-coated vesicles from the membrane. Its role in endocytosis was first discovered in studies of the temperature-sensitive paralytic shibire mutation in Drosophila melanogaster.
Three closely related isoforms exist in mammals:

  • Dynamin 1: expressed exclusively in neurons
  • Dynamin 2: ubiquitously expressed
  • Dynamin 3: primarily expressed in testes

Each isoform has multiple splice variants, suggesting distinct intracellular targets or vesicle budding functions. Common structural domains include:

  • N-terminal GTPase domain
  • Central pleckstrin homology domain
  • C-terminal proline-rich region with multiple SH3-binding sites

These SH3 domains target dynamin to clathrin-coated pits. Dynamin interacts with proteins such as the AP2 adaptor complex and amphiphysin.
