Target Information
AXL is a chronic myelogenous leukemia-associated oncogene also linked to colon cancer and melanoma. The AXL protein is a receptor tyrosine kinase that transduces extracellular matrix signals into the cytoplasm by binding growth factors such as vitamin K-dependent protein GAS6, promoting cell proliferation.
TAM receptors, including AXL, play roles in phagocytosis of apoptotic cells. Mice lacking all three TAM receptors show degenerative phenotypes due to inefficient clearance of apoptotic material, leading to autoimmune symptoms similar to systemic lupus erythematosus.
AXL may also function as an entry receptor for filoviruses and is a marker for Siglec 6+/Axl+ dendritic cells in human blood. Expression of AXL can be upregulated by TLR ligands (e.g., LPS, poly I:C). Soluble AXL, generated by proteolytic cleavage, may serve as a biomarker for inflammation and cancer.
