Target Information
Signal transduction of TGF-beta superfamily members is regulated by Smad proteins. Smads relay signals from the cell membrane to the nucleus and influence gene transcription. Smad4 plays an essential role in TGF-beta-induced transcriptional activation, forming complexes with receptor-associated Smads, SMIF, and BMP4. Increased Smad4 concentration enhances nuclear translocation of this complex. SMIF and Smad4 interact through EVH1/WH1 and proline-rich domains, respectively. Deletion of the Smad4-interacting domain in SMIF prevents TGF-beta-induced nuclear translocation. The human SMIF gene is ubiquitously expressed and encodes a 70 kDa protein.
