Product Specific Information

This antibody detects endogenous levels of TAK1 only when phosphorylated at Ser389.

Target Information

TAK1 (also known as MAP3K7) is a serine/threonine kinase involved in multiple signaling pathways. Initially identified as a TGF-β-activated kinase, TAK1 interacts with TGF-β receptor and TRAF6 to regulate activation of JNK and p38 pathways. It also participates in Wnt, BMP, and activin signaling, and responds to stimuli such as thyroid hormone, osmotic stress, endothelin, and ephrine.

TAK1 contributes to proper wound healing by activating the von Hippel-Lindau tumor suppressor, which represses PDGF-B and integrins β1/β5. It is essential for IKK activation in IL-1, IL-6, IL-18, TNF, CD40, TLR, and RIG-I pathways. Activation of the TAK1–IKK pathway involves TAB2/3 binding, ubiquitination, and autophosphorylation at Thr178, Thr184, Thr187, and Ser192, leading to phosphorylation of IKK-β. TAK1 also represses human telomerase reverse transcriptase, suggesting a role in lifespan regulation.
