Target Information
In T lymphocytes, Ca²⁺ entry following antigen-receptor binding occurs through store-operated Ca²⁺ release-activated Ca²⁺ (CRAC) channels composed of ORAI1 and STIM1. STIM1 acts as a Ca²⁺ sensor and activates CRAC channels by migrating from endoplasmic reticulum (ER)-like sites to the plasma membrane.
STIM2, a related molecule, inhibits STIM1-mediated store-operated Ca²⁺ entry and forms complexes with STIM1, suggesting a coordinated role in Ca²⁺ regulation. At least three isoforms of STIM2 are known.
