Target Information
Estrogen Receptors (ER) belong to the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor acts as a ligand-activated transcription factor that, upon binding to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (EREs) in DNA. Through these interactions, ER can regulate gene transcription by recruiting coactivator or corepressor proteins.
Two main forms exist: ERα (ESR1) and ERβ (ESR2). Alternative RNA splicing generates multiple ERα isoforms (66 kDa, 53 kDa, 47 kDa, 35 kDa). Estrogen receptors are widely expressed and play essential roles in sexual development, reproduction, bone physiology, and are implicated in diseases such as breast and endometrial cancers and osteoporosis.
