Product Specific Information

The 2D1 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells except circulating erythrocytes and platelets. Its cytoplasmic domain exhibits tyrosine phosphatase activity, playing a key role in lymphocyte activation.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometry of normal human peripheral blood cells. Recommended use: 5 µL (0.8 µg) per test (100 µL total volume, 10⁵–10⁸ cells/test).

Important Notes:

  • NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining and should be added before combining antibodies with cells (5 µL/sample).
  • For single-color controls: use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology based on nucleic acid dye structures engineered for minimal spillover and spread impact.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells. It comprises ~10% of lymphocyte surface proteins and is absent from non-hematopoietic tissues. CD45 regulates Src-family kinases, T/B-cell receptor signaling, cytokine receptor pathways, and integrin-mediated survival signaling. Multiple isoforms (e.g., CD45RA) arise via alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions.
