Product Specific Information

This 3G8 monoclonal antibody reacts with human and non-human primate CD16 (low-affinity Fc gamma RIII).
CD16 exists as two isoforms: FcγRIIIA and FcγRIIIB.

• FcγRIIIA: Expressed on monocytes, macrophages, and NK cells.
• FcγRIIIB: Detected on neutrophils as a GPI-anchored form.

Binding of IgG activates signal transduction pathways leading to:

• Antibody-dependent cell-mediated cytotoxicity (ADCC)
• Phagocytosis
• Cytokine release
• Antigen presentation

3G8 recognizes the same epitope as CB16 but distinct from B73.1.
Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometry of human peripheral blood cells (5 µL/test, 100 µL final volume, 10⁵–10⁸ cells/test).

NovaFluor Compatibility Notes:

• Not compatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI).
• Recommended: Invitrogen LIVE/DEAD Fixable Dead Cell stains.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):

• Required for optimal NovaFluor staining.
• Add 5 µL per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor Technology:
Based on Phiton technology utilizing nucleic acid dye structures for engineered fluorescent signatures minimizing spillover and spread.
